What is Spravato?
Spravato is a prescription medicine, used along with an antidepressant, taken by mouth to treat: Adults with treatment-resistant depression (TRD) Depressive symptoms in adults with major depressive disorder (MDD) with suicidal thoughts or actions.
Mental health professionals tend to prescribe esketamine nasal spray for its effectiveness. Patients who are on oral antidepressants can benefit for further treatment augmentation.

Nerve endings rely on compounds known as neurotransmitters. Three main neurotransmitters regulate mood and emotional response in the brain:
- Serotonin, a chemical that regulates emotions like anxiety, happiness, and general mood
- Dopamine, a feel-good chemical that rewards the brain for actions like running (jogger’s high) or eating
- A special Neurotransmitter that affects energy and concentration

This, in turn, leads to an uptick in the production and release of beneficial neurotransmitters. This translates to a considerable improvement of brain function and mood regulation.
In other words, the treatment tackles many of the symptoms that come with depression. Esketamine can also decrease the frequency of suicidal thoughts, which helps with severe depression.
Spravato FAQ
You cannot take Spravato on your own. You will administer Spravato nasal spray yourself under the supervision of a healthcare provider, at a certified Spravato treatment center. Because of the risks for sedation, dissociation, and abuse and misuse, Spravato can only be administered at healthcare settings certified in the Spravato Risk Evaluation and Mitigation Strategy (REMS) Program.
While Spravato is often an effective way to treat treatment-resistant depression, not all patients see noticeable improvements to their condition. Anyone who is not getting the desired results with Spravato should talk to their doctor about alternative ways to treat their depression.
Clinical studies found that patients who took Spravato for treatment-resistant depression typically saw improvements to their symptoms about four weeks after starting their treatments, compared to those who took a placebo nasal spray.
Yes, patients should avoid operating machinery and activities that require them to be alert until they have a full night’s rest after taking Spravato. This means patients need to arrange for transportation to and from the clinic on treatment days. They also have to plan their schedule because their ability to concentrate will be limited for the rest of the day.
